INVITRO PLANT REGENERATION VIA SOMATIC EMBRYOGENESIS IN BAMBOO [Dendrocalamus strictus]

ABSTRACT: The present investigation was carried out to develop protocol for in vitro propagation of bamboo [Dendrocalamus strictus] through somatic embryosis. Four treatments viz. MS+ 2, 4-D 2, 3, 4 and 5mg/L were tried for the initiation, texture and colour of callus. Similarly, four treatments viz. MS+BAP2, 3, 4 and 5mg/L were tested for germination of somatic embryo and no.of shoots initiation. Similarly two concentrations of Hgcl2 were tried for surface sterilization of seed viz. 0.1% [7min.] and 1% [15min.]. Data revealed that seed cultured on solidified MS medium supplemented with 2, 4-D 3mg/L produced embryogenic callus with globular shape embryos. Considerably, the BAP 5mg/L gave better germination of somatic embryos with dark green colour. 15 to 25 shoots were also developed after four weeks of incubation in this treatment. Similarly, 0.1% Hgcl2 for 7 min. was found better for surface sterilization and formed profuse callus, having white and amorphous callus with high frequency of somatic embryo germination. After, development of the plantlets through somatic embryogenesis, same were hardened.
